Nitrosative stress causes dose-dependent growth inhibition of H. capsulatum. H. capsulatum G217B cultures were grown to mid-log phase and treated with a range of concentrations of DPTA NONOate or GSNO. (A) Spectrophotometric readings at OD600 were taken at 0, 2, 5, and 25 h after the addition of DPTA NONOate. The starting OD was set at 100% growth. (B) Colony-forming units relative to the 0-h, 0-mM time point were determined at 0, 2, and 25 h after addition of DPTA NONOate. (C) Colony-forming units relative to the 0-h, 0-mM time point were determined at 0, 2, and 25 h after addition of GSNO. Data shown for A and B are from dose-response experiment 1.
