(A) HEK-293 cells were incubated with 0.1 mM HgCl2 for 10 min before the time course of fluorescence changes in the presence of an inwardly directed ammonium gradient was determined, as described in Figure 2. The same samples were further analysed for pHi changes after treatment with 5 mM 2-mercaptoethanol. (B) Dose–response to mercury inhibition. RhBG transfectants were pre-incubated for 10 min with increasing concentrations of HgCl2 (20, 50, 100, 200 μM) before stopped-flow analysis was performed. Each point represents the mean value of 6 to 8 replicated experiments. IC50 was close to 52 μM.
