Figure S3. Highly abundant hnRNP isoforms and their correlation with gene expression dynamics.
(A) Top 50 genes with the highest abundance of isoforms in our dataset show several hnRNPs. These hnRNPs are expected to bind to intronic splicing silencers (ISSs) or exonic splicing silencers (ESSs) near splice sites, inhibiting spliceosome recognition of the 5′ and 3′ splice sites flanking the alternative exon and preventing their use. The counts of up-regulated and down-regulated isoforms at an adjusted P < 0.05 for each gene are shown in red and blue, respectively. (B) Scatter plot showing the relationship between isoform count and gene expression change (weighted average of isoform log2FoldChange). The x-axis shows the number of isoforms (limited to 70 for clarity), and the y-axis represents the weighted average log2FoldChange for each gene. Gene data points are colored based on whether any isoform is up-regulated (red) in PRMT5-inhibited samples or down-regulated (blue).
