HPLC analysis of tolvaptan sulfate formation in cell lysates from individual SULT-overexpressing cells. Cell lysates were incubated 37°C with 0.1 mM tolvaptan and 0.5 mM [35S]PAPS as described in Materials and Methods. Shown are tolvaptan and tolvaptan sulfate from incubations using 0.4 mg of cell lysate proteins from HEK293/SULT1C3 cells (A and B) and 1.8 mg of cell lysate proteins from HEK293 cells (C and D). Incubations were performed for 1 h with HEK293/SULT1C3 and for 6 h with HEK293 cell lysates.
