Two 733 bp DNA fragments containing the cycB promoter region (nucleotides +236 to –497; the transcription start site at +1) are shown at the top of figure. The seven sites containing the dMyb recognition sequence (AACNG) are indicated. The DNA fragments were immobilized on Sepharose beads and the full-length dMyb protein translated in vitro was incubated with the DNA–Sepharose resin. After washing, the proteins that bound to the cyclin B promoter–Sepharose resin were analyzed by SDS–PAGE. dMyb was detected by autoradiography.
