Loss of FAM136A results in ISR induction
A,B. Analysis of ISR induction upon acute loss of FAM136A in HEK293T (A) and HAP1 (B) cells. Cells were treated with two different sgRNAs to ablate FAM136A. After 3 days the competitive cell fitness assay was started, and after further 6- and 7-days growth on glucose-containing medium, cells were lysed and analysed by immunoblotting for the indicated proteins.
C.,D. Quantification of results shown in (A,B). N = 7 replicates for ATF4 and CHOP in HEK293T. N = 3 replicates for all others. Two-way ANOVA.
E. Model of FAM136A as a disulphide relay substrate involved in IMS proteostasis. See text for details.
