FIGURE 2.

Bar and scatter plots analyzing the effects of different treatments on various cellular parameters. Panels A-G show significant changes in cell viability, LDH release, TNF-ɑ, IL-6, MDA, SOD, and GSH among control, H₂O₂, H₂O₂ with BRD4 overexpression (OE), and H₂O₂ with sh-BRD4 groups. Panel H displays protein levels of Bax, Bcl-2, cleaved caspase-3, and full-length caspase-3 through Western blot images. Panel I illustrates flow cytometry plots for apoptosis across the same groups, with a bar graph indicating apoptosis rates. Asterisks signify statistical significance.

BRD4 regulates H2O2-induced inflammatory responses, oxidative stress, and apoptosis in AEC-II cells. AEC-II cells were assigned to Control, H2O2, H2O2+BRD4-OE, and H2O2+sh-BRD4 groups. (A,B) The viability and LDH release of AEC-II cells from each group. (C,D) TNF-α and IL-6 levels in AEC-II cell supernatant from each group were detected by ELISA. (E–G) MDA, SOD, and GSH levels in AEC-II cells from each group. (H) Bax, Bcl-2, full-length caspase 3, and cleaved-caspase 3 protein levels in AEC-II cells from each group. (I) The apoptosis of AEC-II cells from each group was detected by flow cytometry. Cell experiments were performed in triplicates (n = 3). *P < 0.05; **P < 0.01; ***P < 0.001; ****P < 0.0001.
