Function of SAGA, TFIID, and NuA4 in transcription activation in vitro. (A) SAGA is essential for activation by Gal4-Gcn4. Untagged and Spt7-Flag/Ada1-Flag double-tagged extracts were depleted with anti-Flag beads. Depleted and undepleted extracts were tested in multiround transcription assays either with (+) or without (−) Gal4-Gcn4. A typical experiment in which SAGA purified from a Spt7-Flag extract was added is also shown. The graphs compare absolute levels of transcription under all conditions, and the amount of activation is indicated. (B) TFIID and NuA4 depletion. Untagged and Taf3-Flag/Taf7-Flag- and Esa1-Flag-tagged extracts were assayed as in panel A.
