Figure 2. Degree of genetic overlap between AN and metabolomic markers.

Figure 2.

A. Circos plot depicting the number of significant genetic variants or SNPs overlapping between AN and each of the 249 circulating metabolomic biomarkers determined through conditional/conjunctional FDR analysis, with the height of the bar corresponding to the number of overlapping variants ranging from 0–16, colored by metabolic marker. The dotted line reflects 8 overlapping SNPs, denoting the number of genome-wide significant SNPs found in the original AN GWAS, whereby cFDR analyses with many metabolic markers significantly boosted discovery of genetic variants associated with AN. Two select metabolic markers are labeled in A and further depicted in panel B: Manhattan plots of genome-wide significant SNPs associated with both AN and 2 select metabolic markers, HDL-CE and VLDL Particle Size, with 16 and 15 overlapping SNPs, respectively. Accompanying Venn Diagrams illustrate the genetic overlap between AN and these two markers after bivariate mixture modeling. The estimated number of unique and shared variants are indicated in thousands, with standard deviations in brackets. Bars below the Venn diagrams indicate the concordance rates (“cr”) in direction of effects on the pair of traits, with their genetic correlation (“rg”) listed as a reference. Abbreviations: AN. Anorexia Nervosa; SNPs. Single Nucleotide Polymorphisms; HDL-CE. Cholesteryl esters in high-density lipoproteins; VLDL. Very-low-density lipoproteins.
