Determination of cell populations required for LLO-induced IFN-γ production. Spleen cells were treated with anti-NK (DX5) MicroBeads, anti-CD11b (Mac-1) MicroBeads, or anti-CD11c MicroBeads, followed by depletion of the cells bound to beads by using an MACS LS separation column. The negatively selected cells were adjusted to 5 × 106 cells/ml and stimulated with 20 nM LLO pretreated with cholesterol for 24 h. The level of IFN-γ was determined by ELISA.
