Clonogenic colony-forming activity of Tax-transduced CD34⁺ subpopulations. CD34⁺ HPCs were infected with lentivirus vectors and CD34⁺/CD38⁺/GFP-positive cells and CD34⁺/CD38⁻/GFP-positive cells (10³) were purified by FACS after 72 h and plated in duplicate in 2 ml of MethoCult H4433 (StemCell Technologies, Canada). The absolute number of colonies was determined at 14 days postplating by microscopic analysis.
