Transcription of cytokine genes in THP-1 cells stimulated with FSL-1 and/or ATP or not. THP-1 cells (3 × 10⁶) were stimulated at 37°C for 1 h with 10 nM FSL-1 or not and incubated for 1 h after the addition of 500 μM ATP. The expression of TNF-α, IL-1β, IL-6, IL-8, and IL-10 mRNAs was analyzed by RT-PCR. The identity of the PCR products was confirmed by Southern hybridization with a probe coding for the internal sequence (data not shown). Values in parentheses indicate the intensities of the signals of the mRNAs obtained by densitometric analysis, with the intensity of the signal of the mRNAs of β-actin or various cytokines in the cells in the absence of the stimulators (medium) being taken as 1.
