Removal of chain terminators by the ATP-mediated removal mechanism. 5′-end-labeled primer-templates terminated with tenofovir-PP, ddATP, carbovir-TP, or DXG-TP were incubated with wild-type (WT), K65R, or K65R+M184V RT; and removal of tenofovir, ddA, carbovir, and DXG was evaluated following the addition of 3.2 mM ATP at 0.1, 5, 10, 20, and 30 min (left to right) and subsequent separation through 8 M urea-16% polyacrylamide sequencing gels and exposure to a PhosphorImager screen.
