The PNA-SR oligo specifically alters the bcl-xL to -xS ratio in vivo. (A) HeLa cells were transfected with Lipofectamine and the indicated amounts of PNA-(SR)₈ oligo, the PNA alone or the SR peptide. After 48 h, total RNA was isolated, reverse transcribed, and amplified by PCR. Splicing ratios and error bars have been obtained from three independent experiments. (B) Western blot of protein extracts derived from transfected HeLa cells. Cell lysates were analyzed with a bcl-x specific polyclonal antibody (upper panel) or a polyclonal antibody specific for hnRNP H/H′ as a control (lower panel). The asterisk symbol indicates a non-specific band recognized by the bcl-x antibody.
