H3 histone tails can be phosphorylated on S10 and methylated on K9 in vivo. (A–L) Fixed NIH3T3 cells were labelled with antibodies raised against an H3 peptide carrying a dimethylation on K9 and a phosphorylation on S10. To test specificity, the antibodies were challenged with an excess of the indicated peptides. Panels show mitotic cells strongly labelled by the antibodies. (a–o) Fixed NIH3T3 cells were labelled with the indicated antibodies and DAPI. The presented nuclei were ordered according to the level of H3 S10 phosphorylation and chromatin compaction.
