For D100ms estimates, trajectories of 0.94–16 s long (typically ∼4 s) for cy3-DOPE and 1.5–14 s long (typically ∼10 s) for Gold-DOPE were collected, and their MSD-t plots were linearly fitted between the second and the fourth points (the same as D2–4 in Kusumi et al., 1993). For D3s evaluations, trajectories longer than 3 s were collected, and their MSD-t plots were linearly fitted between 66 ms and 3 s.
D100ms and D3s were observed at a 33-ms resolution (video rate). The SDs include, in addition to the experimental error, the true variations in the diffusion coefficient for individual particles. The distributions for D100ms are shown in Fig. 2.
The hop rates for Gold-DOPE and Cy3-DOPE were calculated using D100ms (median), obtained by SPT of Gold-DOPE and SFVI of Cy3-DOPE (as shown in this table) and the median compartment size determined by SPT (25-μs resolution) of Gold-DOPE (41 nm), based on the relationship: τ = L2/4D100ms ([0.041]2/4/0.19).
