Pictorial representation of lipospermine-DNA binding. (A) Helices are separated due to electrostatic repulsion, (B) as more lipospermine molecules bind to DNA, the acyl chains bound on the same as well as on neighboring helices coalesce together owing to the hydrophobic effect, thereby bringing multiple plasmids together, and (C) despite such a multimolecular coalescence of DNA helices, tight packaging of DNA is precluded due to the steric barrier posed by the acyl chains.
