Effects of LXA₄ analogues on O, NO, and ONOO⁻ formation by LPS-stimulated isolated neutrophils. PMN were incubated with vehicle (control) or one of the LXA₄ analogues (all at 500 nM) for 15 min and then with LPS (1 μg/ml) for 4 h. Superoxide formation was measured as reduction of ferricytochrome c in the culture medium. Intracellular NO formation was monitored using diaminofluorescein (DAF) and is expressed as RFU. DAF fluorescence was reduced 98% by l-NAME. ONOO⁻ formation was assessed as l-NAME-inhibitable oxidation of DHR 123 to rhodamine and is expressed as RFU.
