Wild-type and sac3Δ cells were spotted in 10^–1 dilutions onto YPD plates and growth was analyzed after 3 days at the indicated temperatures. (B) Deletion of SAC3, NUP1 or NUP60 causes an mRNA export defect. Wild-type, sac3Δ, nup1Δ, nup60Δ, nup2Δ and mex67-5 cells were grown at the indicated temperatures and localization of poly(A)⁺ RNA was assessed by oligo(dT) in situ hybridization. DNA was stained with 4′,6-diamidino-2-phenylindole (DAPI).
