Figure 1:
For each of the 126 metastatic NSCLC cases in the TRACERx cohort (6), we used PairTree to obtain an alternative tree to that reported by CONIPHER on the same set of mutational clusters employed by CONIPHER in the original study. a. First, for each case, all PairTree produced trees were evaluated and the top 10 trees according to their reported likelihood were identified: the left panel depicts the top 10 trees for case CRUK0412 (out of 1795 trees sampled by PairTree) - on each node, the number of mutations in the cluster assigned to that node is provided. The right panel depicts the discordance between each pair of trees - defined as the omltd normalized by the total number of mutation labels. b. Next, each of the top 10 PairTree trees for each case is aligned with the CONIPHER tree, and the one with the lowest discordance is identified. The panel shows for case CRUK0412, the discordance between each PairTree tree and the CONIPHER tree. Since, with respect to its reported likelihood, tree 2 has the lowest discordance, it is identified as the PairTree alternative for this case.
