Figure 1: Translation of a peripheral nerve-specific oxazine fluorophore platform into BBB-permeable scaffold for white matter tract imaging.
(a) Prior asymmetric oxazine fluorophores (LGW01–08, LGW05–75) developed for peripheral nerve imaging, established a blood-nerve barrier-permeable NIR fluorescent nerve-sparing fluorophore platform. (b) Rational scaffold engineering in the present work introduced tertiary/tertiary amine termini, asymmetric lipophilicity balance, and morpholino/tetrahydroquinoline features to support BBB permeability while preserving NIR fluorescence emission, yielding novel 700 nm CNS-targeted oxazine scaffolds for WMTs imaging.
