General chemical structures for class A and class B fluorophores, representing all 18 fluorophores. The scaffold was engineered to introduce tertiary/tertiary amine termini and an asymmetric amphiphilic architecture through a morpholino containing side and a more hydrophobic dimethyl tetrahydroquinoline containing side, while auxochrome nitrogen substitution was varied to further tune BBB permeability and WMTs-specific imaging properties.
