iRD21 forms an aggregate under the acidic conditions. A, The extract (20 μg of protein) from the leaves of 35-d-old Arabidopsis plants was incubated at pH 8.0, 7.0, 6.0, or 5.5. The formed insoluble aggregates were subjected to SDS-PAGE followed by immunoblot analysis with anti-RD21 antibodies. The extract (20 μg of protein) before incubation is also shown on the blot (input). B, The extract (100 μL) from the vacuoplasts of the transformant BY2/RD21 cells was incubated at pH 8.0 or 5.5. The formed insoluble aggregates (ppt) and 20 μL of supernatant (sup, × 1/5) were subjected to SDS-PAGE followed by immunoblot analysis with anti-RD21 antibodies.
