As³⁺ induces accumulation of GADD45α mRNA. (A) The levels of GADD45α mRNA and GAPDH mRNA were determined by RT–PCR using cell lysates from the cells treated with 20 µM As³⁺ for the indicated times. The sizes of PCR products were estimated by the DNA molecular marker (M) in base pair (bp). The possible alternatively spliced fragments of GADD45α mRNA were indicated as ‘b’ and ‘c’. Data are representative of at least three experiments. (B) Fold changes in GADD45α expression were determined by real-time RT–PCR. BEAS-2B cells were treated with 20 µM As³⁺ for the indicated times and fold induction of the GADD45α mRNA was measured by real-time RT–PCR as described in ‘Materials and Methods’.
