Toxin A (100 ng/ml) induced Bid activation at 6 h by a caspase-independent mechanism. Pretreatment with inhibitors of caspase (Casp) 8 (40 μM) and caspase 1 (40 μM) and general caspase inhibitor (z-VAD; 20 μM) did not prevent the cleavage of Bid induced by toxin A. T84 cells were preincubated for 1 h with the respective caspase inhibitor, followed by incubation with toxin A. Bid cleavage was analyzed by immunoblotting in cellular lysate of T84 cells incubated with toxin A for 6 h.
