Expression of the HSV-1 capsid proteins in insect cells. Sf9 cells (1 × 106 cells in 35-mm dishes) were infected with recombinant baculoviruses at an MOI of 5 PFU/cell. The cells were harvested at 24, 48, 72, and 96 h after infection for Western blot analysis (B to D) or pulse-labeled with [35S]methionine for 2 h (A). Lysates were analyzed by SDS-PAGE followed by autoradiography for VP5 (A) or transferred to membranes followed by incubation using antibodies to the different capsid proteins (UL26C [B], UL38C [C], and R2421 [D]).
