Analysis of PyLT acetylation in vivo. Cell extracts from NIH 3T3 cells transiently cotransfected with expression vector for PyLT and an expression vector for Gal4 fusion PCAF were immunoprecipitated with rabbit polyclonal anti-acetyl-lysine antibody (α acetyl lysine). The immunoprecipitates (upper panel) were analyzed for PyLT by Western blotting by using anti-PyLT antibody (α PyLT). Expression of Gal4 fusion PCAF and its mutants (middle panel) and expression of PyLT transfected (bottom panel) also were analyzed in one eightieth of whole-cell extract (WCE) by Western blotting by using anti-GalDB (α GalDB) and anti-PyLT (α PyLT) antibodies.
