Divalent cation-dependent binding of TSP1 to TSG-6. ¹²⁵I-TSP1 binding to different amounts of full-length TSG-6Q (panels A, C) or Link_TSG6 (panels B, D) coated onto wells of a microtiter plate was tested by the solid phase assay. Temperature dependence was examined by adding ¹²⁵I-TSP1 (0.5 μg/ml, 50 μl/well) and incubating at 4°C, 22°C, or 37°C in humidified atmosphere at pH 7.4 for 3h. The assays were performed in the presence of Ca²⁺ and Mg²⁺ (Solid lines, panels A, B) or without Ca²⁺, Mg²⁺ and in the presence 5 mM EDTA (dotted lines, panels C, D). Background values were subtracted from each data point (66.0 ±15.6, 49.0 ±3.4, 126.1 ±10.9). Results (mean cpm ± S.E.M.) shown are representative of two independent experiments performed in triplicate.
