Lanes with an asterisk show sample plus mollicute primers (Mo) alone. All other lanes are controls, with PrP primers (PrP) to test for integrity of the sample DNA or mollicute primers plus spikes of 1 pg S. mirum DNA (s1), 0.1 pg S. mirum DNA (s0.1), or 1 pg M. arginini DNA (m1). No products were detected in any of the unspiked samples after first-round PCR with mollicute primers. (B) Second round of nested PCR (30 cycles).
