Immunofluorescence analysis of DC subset distribution in human Peyer's patches. Human terminal ileum tissues were embedded in OCT medium, and 6- to 8-μm frozen sections were stained with antibodies against MHC class II (HLA-DR/DP/DQ) (green) and CD11c (red) (A and B) or IL-3R (CD123; red) (C and D). The subepithelial dome region (B) and the interfollicular regions (D) containing CD11c+/MHC class II+ or IL-3R+/MHC class II+ DCs, respectively, are shown at higher magnifications. Images were captured using either 10× (A and C) or 40× (B and D) objective lenses. No background staining was detected when mouse IgG was used as a control (data not shown).
