(a) HEK293 cells were either mock infected or infected with T3A, T1L, or UV-inactivated T3A at an MOI of 100 PFU per cell. mRNA was collected at 24 h postinfection and analyzed for chk1, wee1, and GADD45 transcripts by RT-PCR to confirm microarray analyses and also to assess the importance of replication competence for the observed changes in gene expression. β-Actin was used as a control in each RT-PCR to ensure matched total mRNA and equivalent amplication conditions for each experimental sample.
