The Cas12k–TnsB–TnsC–TniQ complex directs the integration of donor DNA 60–66 bp downstream of the protospacer-adjacent motif (PAM). The inserted sequence, flanked by left-end (LE) and right-end (RE) transposon terminal repeats, results in a 5-bp target-site duplication upon integration.
