FACS analysis of the effect of α and β RNAi knockdown on the mitochondrial membrane potential. Analysis of 2.5 × 106 cells/ml each of wild type (WT) and RNAi-induced cells stained with a 2.5 μM concentration of the membrane potential sensitive MitoTracker Red is shown. CMXRos (red trace of each panel) and treated with 50 μM CCCP prior to MitoTracker staining (green trace of each panel) were analyzed with the FACSCalibur flow cytometer to monitor the changes in fluorescence intensity.
