Southern blot analysis of four BoHV-1 infectious BAC clones following the replacement of the gene coding for glycoprotein E (gE) with a kanamycin resistance gene (Kan^r). Viral genomic DNA was isolated from cells infected with modified virus. DNA was digested with either BamHI or HindIII, resolved on a 1% agarose gel by FIGE, and transferred onto nylon membranes. Blots were hybridized with a DIG-labeled probe specific for either gE or Kan^r. Standard molecular sizes (in base pairs) are given. (A) Southern blot hybridized with a probe specific for the gE gene. (B) Southern blot hybridized with a probe specific for the Kan^r gene.
