Digestion of supercoiled (SC) DNA by HinP1I in solution. (A) Each 5 µl reaction in the New England Biolabs (NEB) buffer 3 (50 mM Tris, pH 7.9, 10 mM MgCl₂, 100 mM NaCl and 1 mM DTT) contains 0.16 µg of pUC19 DNA and 1 ng of HinP1I (a 1:4 molar ratio of HinP1I to pUC19 DNA). The reaction was carried out at room temperature for the indicated times. (B) Same as (A), except the NEB enzymes (EcoRI and BamHI) were used.
