Protein processing and maturation profiles in cell lysate and supernatant virions of NL4-3 and molecular clones carrying variant and mutant stem-loops. (A) For sodium dodecyl sulfate-polyacrylamide gel electrophoresis, COS-7 cells were transfected with recombinant clones and metabolically labeled with [³⁵S]methionine-[³⁵S]cysteine. Proteins were immunoprecipitated with anti-HIV human immunoglobulin G and analyzed by 5 to 15% gradient gel electrophoresis and fluorography. (B) For Western blotting, proteins from supernatant virions were reacted with anti-p24 polyclonal and anti-RT monoclonal antibodies. Shown is a representative blot from three separate experiments.
