Fluorescence time course for manganese binding. Apo-MnSOD (50 μg/mL) in 20 mM MOPS, pH 7 was equilibrated at 45°C in a thermostated cell holder as described in Experimental Procedures and MnCl₂ was added (to a final concentration of 10 μM) to initiate the reaction. A theoretical fit to the data based on two-exponential relaxation is shown (open line).
