Cell lysate (C.L.) and crude membrane (M) samples from MDTF cells infected with retrovirus carrying FLVCR1, FLVCR2, or hybrid FLVCR1/FLVCR2 constructs were analyzed for protein expression. The loading controls for actin and Na⁺ K⁺ ATPase are shown. (A) These proteins were tagged with V5 epitope and detected using monoclonal anti-V5 antibody and rabbit anti-mouse HRP antibody. Relative surface expression of proteins was determined by using Image J software (see Materials and Methods). Surface expression of proteins is relative to expression of FLVCR1. (B) These proteins were tagged with an HA epitope and detected using anti-HA HRP antibody. Surface expression is relative to the expression of 2-1^TM1.
