J774.1 cells were maintained in DMEM containing 10% FBS and then infected with bacteria at an MOI of 10:1. Two hours after infection, unphagocytosed organisms were removed. Mycobacterium-infected macrophages were maintained in DMEM containing 10% FBS in the absence and presence of IFN-γ (100 U/ml) plus LPS (1 μg/ml). Infected macrophages were terminated at 1 h and 72 h to determine intracellular growth of bacteria inside J774.1 cells.
