Phosphorylation of ZEBRA by different PKC isoforms in the absence (−) and presence (+) of bisindolylmaleimide I. In vitro phosphorylation reactions by different PKC isoforms (α, β1, β2, γ, ɛ, ζ, and μ) were performed using 250 ng of His-tagged ZEBRA and [γ-³²P]ATP. The inhibitory effects of 0.5 μM bisindolylmaleimide I on different PKC isoforms were examined by the degree of autophosphorylation of each isoform in the absence or presence of the inhibitor. The reaction products were separated by SDS-10% PAGE, dried, and autoradiographed.
