Incorporation of [α′-32P] UTP into RNA using isolated nuclei preparations was carried out as described in Materials and Methods. DNase treatment was at 28°C for 30 min prior to addition of NTPs using 5 units of RNase-free DNase. RNase treatment was at 28°C for 30 min after completion of synthesis using 100 μg/ml RNase A.
