(A) Intracellular concentrations of chimeric R228, substrate, and noncleavable substrate snoRNAs with a G+1A mutation in yeast grown in media with 2% glucose or with 2% sucrose and galactose concentrations ranging from 0% to 2%. (B) An Eadie-Hofstee plot of the chimeric R228 snoRNA concentration dependence of rates of chimeric substrate snoRNA cleavage in yeast nuclei gives a kcleav value of 0.037 ± 0.004 min−1 and a KMS value of 1.0 ± 0.2 μM.
