LeishIF4E-1 binds only to m⁷GTP and not to TMG. The soluble fraction of E. coli BL21 cells induced by IPTG (lane a) was applied to a TMG-Sepharose column. After collection of the flow-through fraction (lane b), the column was washed with equilibration buffer (lane c) and eluted with the same buffer containing 200 mM NaCl (lane d). The same extract (lane a) was loaded over m⁷GTP-Sepharose, the flow-through fraction was collected (lane e), and the column was washed with equilibration buffer (lane f) and eluted with the same buffer containing 200 mM NaCl (lane g).
