β-Galactosidase activity correlates with the stability of the loop–loop interaction. The β-galactosidase activity is normalized relative to the activity of X1–Y1. Error bars indicate standard deviation. The constructs X1a–X1c and Y1a–Y1c were assayed against their respective cognate partner (dark gray). As a control, the X and Y constructs were tested against the corresponding 1 construct: X1 for the Y RNAs (light gray) and Y1 for the X RNAs (white). Sequence, stability, and melting temperature at a concentration of 10−5 M of the different constructs are indicated.
