Prior to the import reaction, chloroplasts (15 µg of chlorophyll) were incubated with deamino-NAD and HAR for 15 min in the dark. Import was performed for 4 min at 25°C. The chloroplasts were recollected and analysed by SDS–PAGE and autoradiography. The mature peptides were quantified using a phosphoimager. Means of at least five independent experiments are given.
