Overexpression of SCAMP2, particularly mutant B, does not noticeably affect the distribution of plasma membrane SNAREs, syntaxin 1, and SNAP25. PC12 cells transfected with myc-tagged mutant B under the same conditions as in Figure 6, C–E, were examined by double-label immunofluorescence at the whole-cell level and as plasma membrane sheets prepared by light sonication. Representative images of plasma membrane sheets stained with antibodies against myc epitope plus syntaxin 1 and myc epitope plus SNAP-25 are shown and illustrate the similar and partially overlapping distributions of SCAMP2 with the SNAREs.
