Newly synthesized Pma1p does not acquire detergent resistance in elo3Δ at 37°C. elo3Δ pep4Δ (YRS1550) cells were incubated at the indicated temperatures for 15 min, pulse-labeled for 5 min, and chased for 15 and 60 min. Cell lysates (T) were then extracted with Triton X-100 and separated into a soluble (S) and insoluble (P) fractions. Pma1p was immunoprecipitated and analyzed by SDS-PAGE and fluorography.
