(e to g) To test the ability of cells to form multiple layers (focus formation capacity), 106 cells (passage 20) representing Cdk4+/+ (e), Cdk4+/R24C (f), and Cdk4R24C/R24C (g) MEFs were plated in 10-cm-diameter dishes and grown for 2 weeks. Medium was changed every 3 days, and at the end of 2 weeks, plates were stained with Giemsa stain to determine the focus formation capacity.
