The template strand (05) and the release strand (06) were chemically synthesized in both ssRNA and ssDNA and annealed to form a dsRNA or a dsDNA substrate. The 5′ end of the release strand was first labeled with [γ-32P]ATP before annealing. The numbers above the substrate represent the position of the nucleotide in the HCV genome.
