C terminus of Cuf1 is required for copper-dependent cytoplasmic localization. The cuf1Δ strain of S. pombe JSY17 was transformed with plasmids that encode GFP, ¹Cuf1⁴¹⁰-GFP, ¹Cuf1³²²-GFP, ¹Cuf1¹⁸⁰-GFP, or ¹Cuf1¹¹⁶-GFP proteins. Cells were grown to mid-logarithmic phase in medium supplemented with 5 μM thiamine. Thiamine was withdrawn from cell cultures, and then cells were grown for 4 h in CuSO₄ (100 μM) or BCS (100 μM). Cells were analyzed by fluorescence microscopy for GFP (GFP). Cells were also examined by Nomarski microscopy for cell morphology (Nomarski).
